Enzyme assisted extraction of oil from rapeseeds with minimal addition of water

Abstract: An enzyme assisted extraction for increasing the recovery of oil from flaked rapeseeds was investigated, with application of enzymes directly onto the seeds i.e., with minimal water addition ( < 1%). A combination of pectinases and cellulases were evaluated. The enzyme application method was evaluated by determining the variation of the water content and water activity of subsamples of seeds from the same bath, from which it was concluded that the enzymes were evenly distributed. It was evaluated if the enzymes had the desired effect of breaking down the polysaccharide structure of the rapeseeds by measuring the content of reducing sugars by a DNS assay. The use of pectinases resulted in a significant increase in reducing sugar content while the cellulases resulted in no significant difference compared to a control without enzymes (P < 0.05). Oil was extracted from rapeseeds treated with a 1:1 mixture of pectinase and cellulase at 0.05, 0.2, or 0.5 g enzyme per 100 g seeds using a continuous screw-press, and oil recovery was determined from the residual oil content in the press cake as analyzed by Soxhlet.
The oil recovery was increased from 60.1 % to 65.0 % by applying an enzyme dosage of 0.5 % (P < 0.05). The oil was analyzed by electron spin resonance spectroscopy to determine if any difference in the oxidative stability was apparent for the oil extracted with enzymes, where no significant difference was found compared to oil extracted without enzymes (P < 0.05). The work was done in collaboration with Siccadania AS and Dansk Landbrugs Grovvareselskab (DLG) and it is supported by the Danish Green Development and Demonstration Program (Grønt Udviklings- og DemonstrationsProgram; GUDP).
