Microwave aided extraction (MAE) method optimization to produce canolol from canola meal

Abstract: The de-oiled canola meal contains wide array of phenolic compounds with sinapine and sinapic acid the predominant ones. Under thermal treatment, sinapine undergoes hydrolysis to sinapic acid, while the latter then converts to canolol via decarboxylation. Canolol can be used as a natural antioxidant to prevent the lipid oxidation and improve the shelf life of food products. Microwave-assisted solvent extraction (MAE), as a green extraction method, has received considerable attention in recent times. The application of MAE, incorporating green extractants could enhance the yield of canolol as its formation is primarily dependent on temperature. In the current study the effects of different pre-treatment temperature-time combinations of 140, 150, 160, and 170 ℃ for 5, 10, 15, 20 and 30 minutes on the extraction of canolol and other canola endogenous phenolic compounds were evaluated using the MAE. Total phenolic content (TPC), total flavonoid content (TFC) along with metal ion chelation (MIC) and DPPH radical activity of the different extracts were assessed. The results confirmed that extractability of canolol was optimized for methanol at 151 ^oC and for ethanol at 170 ^oC with pre-treatment times of 15.43 min and 19.31 min respectively. Further, statistical analysis found there was a strong positive correlation between TPC and TFC (p < 0.05). However, no significant correlation was observed between MIC and DPPH. The results obtained with MAE confirmed its use as a novel and versatile instrument for enhancing the yield of canolol. Improvements in the antioxidant activity of the different extracts further established the efficacy of the current method for isolating canolol and important natural phenolic derivatives for utilization by the nutraceutical industry.
