Iron-based amorphous alloy as potential support material for the immobilization of Rhizopus niveus

Abstract: Lipases are among the top-rated industrial enzymes due to their multifunctionality, as they accomplish both synthesis and hydrolysis reactions. However, as with any other proteins, they are vulnerable to changes in operating and storage conditions. Therefore, immobilization of enzymes on a solid support is one of the robust approaches to enhance their stability and recyclability. In this study, the immobilization of Rhizopus niveus lipase (RNL) on iron-based amorphous alloy (Fe-AA) was conducted to obtain stable biocatalysis reaction. Fe-AA are promising materials in the field of biomaterials with superior mechanical performances, corrosion resistance, unique thermal properties, and acceptable biocompatibility. The activity was evaluated using calorimetric assay with pNPP (p-nitrophenyl palmitate) as a substrate for lipase hydrolysis. At the dose of 20 % wt/vol, 350 rpm, 40 °C and 3 h incubation, protein concentration was observed to be reduced in the solution, indicating the adsorption of the RNL into the Fe-AA. Further analysis shows that the immobilization efficiency of the RNL@Fe-AA was 18.3 % after 3 h. The relative activity of the RNL@Fe-AA increased to 113 % under optimum conditions (pH 7, 350 rpm, 40 °C and 3 h incubation time) in comparison with free enzyme (control sample, set as 100 %). The study presents a new magnetic support material for enzyme immobilization that improves enzyme stability, particularly in biochemical processing applications such as hydrolysis. Although the immobilization shows not a promising result, it can be expected that modification or functionalization with co-solvents would enhance the efficiency of the adsorption of RNL to Fe-AA.
