Eutectic solvents and nanocellulose for tuning of enzymatic properties

Abstract: The biotechnological industries all over the world are currently looking for ways to improve enzyme productivity and create new techniques to extend their shelf life. Generally, when it comes to the growth of numerous chemical industries, such as food, bioenergy, or pharmaceutical; enzyme immobilization is extremely important. For use in non-aqueous mediums, immobilization is frequently the key to increasing an enzyme operational efficacy in industrial operations as free enzyme are often susceptible to the denaturation. To achieve this goal, Candida Rugosa lipase (CRL) was immobilized with three different groups of natural deep eutectic solvent (NADES) and two types of nanocellulose (NC), respectively. The immobilized CRL are more durable and resistant to environmental changes than free enzymes in solution. Noteworthy, the NC extracted from almond shells synthesized by perchloric acid (NC-PA) and methanesulfonic acid (NC-MSA) demonstrates higher relative activity up to 480% and 316%, respectively when subjected to glucose based NADES (ChCl:sucrose:water in molar ratio 4:1:4). It also provides a strong framework for enhancing enzyme availability to substrate as demonstrated by scanning electron microscopy (SEM) studies which revealed significant hydrogen bonding interactions in immobilized lipase. As a result, lipase immobilization in conjunction with an innovatively modified biocatalyst (NC-NADES) can improve the condition of the enzyme and offer new perspectives to the oil industrial sector for the treatment of oil contaminants to fulfil world commercial demands.
