Abstract: The role of activated carbon (AC) as an immobilization material for enhancing lipase-catalyzed reactions in an aqueous solution with deep eutectic solvent (DES) as a co-solvent was investigated in this study. The effects of incubation time, reaction temperature and water content on lipase activity were studied. The enzymatic activities of Amano lipase from Burkholderia cepacia (AML) and lipase from the porcine pancreas (PPL) were used to investigate the optimum conditions. The results showed that PPL has better immobilization on AC and higher enzymatic activity than AML. DES composed of glycine and sodium hydroxide (1:1 molar ratio) with AC to further enhanced enzymatic activity. Under optimized conditions (2 h, 50 ºC, and 80 % water content), the immobilized PPL-DES-AC and PPL-DES gave a maximum relative activity of 285% and 260.3 respectively, compared to the free CRL 100%. The kinetic parameters VMAX of the PPL immobilized on DES-AC was also estimated to be 0.39 and 2.77 mM min−1, and the KM was calculated as 0.74 and 4.8 mM, respectively. The kinetic parameters demonstrated that the immobilization of AC enhanced the activity of PPL in a DES medium.
